PEG MGF, also known as PEGylated Mechano Growth Factor, is a polyethylene glycol conjugated synthetic peptide derived from the C-terminal E-domain of IGF-1Ec, an alternative splicing isoform of insulin-like growth factor 1. Native unmodified MGF is locally expressed within skeletal muscle tissue when muscle suffers mechanical loading, strain or injury. However, the native MGF peptide exhibits extremely short circulating half-life, only several minutes, which greatly limits its application in systemic preclinical research. PEGylation modification is adopted to covalently attach a methoxy polyethylene glycol chain to the N-terminal of the MGF peptide backbone. This PEG chain forms a protective hydration shell around the peptide molecule, shielding it from rapid enzymatic degradation and renal clearance, which drastically extends its effective duration in biological models.

The primary research target of PEG MGF lies in the activation of muscle satellite cells, which are dormant resident stem cells located between muscle fiber and basement membrane. Upon stimulation by PEG MGF, these quiescent satellite cells get activated, start proliferation and differentiation, fuse into damaged muscle fibers, and trigger the cascade of muscle protein synthesis.
